After receiving the fibroblasts, they need to be stabilized in the incubator for about 4 hours and then cultured or passaged according to the cell density, changing the medium. The passaging operation follows the following steps:

1. Aspirate the medium in the original cell culture flask, rinse the cells twice with PBS buffer, and add 1~2 ml of 0.25% EDTA for trypsin digestion (pay attention to the digestion time, usually controlled within 1~2min).

2. Observe the digestion under the microscope. When the cell edge shrinks and adheres to the wall (digestion is not recommended until the cells float), remove the trypsin, add 6~8ml of complete medium, gently blow the cell layer, and try to blow off and blown away the cell layer.

3. Take out part of the cell suspension and transfer it to a new cell culture flask, add appropriate complete medium, and cultivate in an incubator.

4. Pay attention to the change of the pH value of the medium, change the medium regularly, and repeat the subculture or freeze storage when the cell density reaches 70-80%.

The main role of cell culture flasks in cell passaging is to store cells and provide an adherent growth environment for cells. It should be noted that if the cell wall is broken or leaking after receiving the cells, it means that the cells have been contaminated, and the supplier should be contacted in time.
