Application of erlenmeyer shake flask in Jurkat cell culture

Fri Aug 19 15:23:04 CST 2022

The erlenmeyer shake flask is a special culture container for suspension cell culture, and can also be used to prepare, mix and store various media. This culture consumable is used when culturing Jurkat cells.

The Jurkat cell line is derived from the peripheral blood of a 14-year-old boy and is a suspension cell. Jurkat-derived cell lines lacking certain genes are already available in cell culture banks. Immortalized human T lymphocyte lines mainly used to study acute T cell leukemia, T cell signaling, and various chemokine receptors susceptible to viral entry, especially HIV expression. It has a very wide range of applications in biological research, such as the application of Jurkat cells to study the M1-RNA of ribonuclease P, and the study of the M1-RNA of anti-MHC class II transcriptional activator (CIITA) to inhibit the expression of MHC class II molecules on the cell surface.

When culturing Jurkat cells in erlenmeyer shake flasks, RPMI1640 medium, 10% FBS is required; the temperature is controlled at 37°C, 5% carbon dioxide, PH value 7.2-7.4, aseptic constant temperature culture. Wipe and disinfect with 75% alcohol before transferring to the cell ultra-clean bench, take out the cell cryovial from the liquid nitrogen tank, immediately place it in a 37°C water bath and shake the cell cryotube quickly to thaw it quickly. Then, after centrifugation, pipetting and mixing, etc., it was placed in a cell incubator for cultivation.

Cells are particularly sensitive to the environment. When culturing Jurkat cells in erlenmeyer cell shake flasks, personal hygiene should be done well, sterile reagents should be used, and aseptic operation principles should be followed to avoid introducing bacteria and affecting cell growth.

The FAI climbed 5.9 percent year-on-year in the first 11 months of 2018, quickening from the 5.7-percent growth in Jan-Oct, the National Bureau of Statistics (NBS) said Friday in an online statement.

The key indicator of investment, dubbed a major growth driver, hit the bottom in August and has since started to rebound steadily.

In the face of emerging economic challenges home and abroad, China has stepped up efforts to stabilize investment, in particular rolling out measures to motivate private investors and channel funds into infrastructure.

Friday's data showed private investment, accounting for more than 60 percent of the total FAI, expanded by a brisk 8.7 percent.

NBS spokesperson Mao Shengyong said funds into weak economic links registered rapid increases as investment in environmental protection and agriculture jumped 42 percent and 12.5 percent respectively, much faster than the average.

In breakdown, investment in high-tech and equipment manufacturing remained vigorous with 16.1-percent and 11.6-percent increases respectively in the first 11 months. Infrastructure investment gained 3.7 percent, staying flat. Investment in property development rose 9.7 percent, also unchanged.
