How to observe cells in cell culture flasks

As an easy-to-use cell culture container, cell culture flasks are very frequent in the laboratory and are mainly used to culture adherent cells. When culturing cells, we need to observe the state of the cells and judge their growth. Then, how should we observe it?

First of all, observing the state of the cells can be done by eyes or microscope. When we take out the cell culture flasks from the incubator, we can judge the growth of the cells by the color of the medium, the concentration, and the state of the cells.

1. Look at the color of the culture medium. Most culture mediums have acid-base indicators, with acidic color showing yellow and alkaline color showing purple. If the pH value of the medium changes greatly, consider whether the medium is contaminated, or the cells grow too densely, or the cells die too much, or there is a problem with the regulation of CO2 concentration in the incubator.

2. Look at the concentration of the medium. If the medium becomes turbid, it means that the medium is contaminated or the cells are growing too densely.

3. Look at the cell state. Cells in good growth condition can be seen under the ordinary microscope, the cell transparency is large, the outline is unclear, only with the phase contrast microscope observation, you can see the fine structure of the cell. Few intracellular particles, no vacuoles, clear cell membrane, clear and transparent culture supernatant, can not see the suspended cells and debris; cell dysfunction, the contour of the enhanced cytoplasm often appear vacuoles, lipid droplets and other granular material, the gap between the cells to increase the cell morphology can become irregular, or even lose the original features.

In summary, we can judge the growth status of cells by observing the color, concentration, and cell status of the medium in the cell culture flasks. The culture conditions of each experiment are different, and only by always mastering these conditions can we make timely adjustments and do a good job in the subsequent experiments.
