培地は細胞培養の過程で不可欠な栄養素です。細胞培養にセルファクトリーを使用する場合、通常、培地交換操作が必要です。これにより、増殖プロセス中に細胞によって生成される代謝廃棄物を除去し、新鮮な培地を補充して細胞の増殖と再生を改善できます。
セルファクトリーの培地を交換するには、まず必要な機器を準備します。アルコール散水缶に75％の医療用滅菌アルコール、PBSバッファー、血清含有培地、液体トランスファーカバー、トランスファーチューブ、廃液タンクなどを充填できます。 。次に、すべての機器を超清浄ベンチに置き、滅菌のためにUV照射をオンにします。培養細胞が温度に敏感な場合は、血清を含む培地ボトルを37°Cの水浴に入れて、培地の温度が37°Cになるようにしてから、UV用の超清浄ベンチに移します。細胞培養の合間には、滅菌とUV照射も約30分必要です。
紫外線照射滅菌後、オペレーターは滅菌服、滅菌手袋、マスクを着用し、細胞工場を取り出し、表面に75％のアルコールを噴霧しました。消毒用のアルコール散水缶を使用して、それを超清浄ベンチに移し、蓋を開け、液体移送キャップと移送チューブを接続して培養培地を輸出しました。新鮮な血清含有培地を添加し、細胞の付着面からではなく側壁から添加するように注意し、細胞が洗い流されないように静かに動かします。培地を追加した後、蓋を覆い、消耗品の表面に印を付け、培地の交換時間、セルの種類、オペレーター、およびその他の情報を示します。インキュベーターに入れる前に、表面にアルコールを再度スプレーし、手術台を片付け、超清浄テーブルをアルコールで拭き、廃液やゴミをきれいにします。
細胞は環境に敏感です。細胞工場で培地を交換する場合は、培地交換の過程でさまざまな細菌に感染して細胞増殖に影響を与えないように、無菌操作に注意を払う必要があります。
The FAI climbed 5.9 percent year-on-year in the first 11 months of 2018, quickening from the 5.7-percent growth in Jan-Oct, the National Bureau of Statistics (NBS) said Friday in an online statement.
